The Rare Diseases Research Unit (RDRU) of Pfizer is committed to the development of new therapies to address the unmet medical needs of those individuals with rare and neglected diseases. Recently, the RDRU established Bamboo Therapeutics in Chapel Hill, North Carolina in order to develop novel gene therapies using AAV. The RDRU is seeking to recruit a laboratory scientist to support Bamboo's goal of advancing development of novel capsid for targeted gene transfer, immune response, and efficient and persistent tissue specific transgene expression. The candidates will be expected to oversee and manage computational problems related to structural biology with respect to virus host interactions, develop algorithms to examine the interaction of amino acids of the capsid with the adaptive immune response. Model changes in the capsid with transduction changes seen in target organs of animal models. The successful candidate will contribute directly to projects in the lab that seek to uncover new avenues for therapeutic development. This position involves working closely with a multidisciplinary project team to design and execute multi-day experiments in addition to performing complex data analysis. The aim is to integrate in vitro and in vivo model studies with human biology to inform gene therapy discoveries. The candidate must be a strong experimentalist with the ability to lead from the bench while playing a key role in hypothesis generation. Our lab develops emerging targets applicable to Duchenne Muscular Dystrophy and other rare diseases rooted in human biology and genetics. We are seeking new members of our team that focuses on innovative solutions to the challenges of rare disease research sphere. These scientist positions will focus primarily on the use of AAV.

The key responsibilities for this role is the analysis of capsid structural changes on in vitro as well as in vivo targeting in mouse models with respect to the molecular interaction between the capsid and the target cell. This position will focus on the generation of new vectors (both in vitro and in vivo) in addition to the development and validation of assays to power gene therapy discovery campaigns. Another key responsibility will be in the design and execution of studies to understand the specific role of receptor-virus interactions, virus-immune response interactions, and vector transgene transcription/translation interaction in both normal and diseased tissue/cell lines. This requires comprehensive understanding of the molecular biology of parvovirus DNA replication, transcription, persistence, and immune response after virus infection. Critical to the success of this position is a fundamental understanding of viral structure-function interactions and the role these peptides may play in successful infection, transduction, and template persistence. It is also preferred that the candidate have a fundamental understanding in vector DNA template genomic interaction related to persistence and vector chromosome insertion.


+ PhD with 5+ years or Master's Degree with 9+ years of experience in a biochemical, molecular, or cellular-related bioscience discipline with 5+ years of laboratory experience including degree research

+ Candidate must have 3+ years experience working with AAV biology and AAV vectorology.

+ Candidates must have 3+ years of independent lab research experience working with AAV vectors.

+ Candidate must have extensive experience managing a research team focused on project-specific goals as well as able to facilitate joint collaboration between research teams.

+ Candidate must have prior experience working in an industry environment.

+ Expertise in publications, presentations, and managing subordinate research projects is necessary.

+ Proficiency in Excel, Word, PowerPoint and scientific graphing software.

+ Experience with statistical analysis software such as R and ability to perform basic scripting programming is desirable.

+ Experience with PyMol and other structural programs for analyzing 3D data are required.


+ Demonstrated abilities with molecular biology, biochemistry and imaging techniques to study vector transduction in vitro and in vivo

+ Track record of hypothesis-driven research that has employed molecular, biochemical, and cellular methods

+ Experience with the development of viral vectors for efficient transduction in vivo in rodent models

+ Team player who can communicate effectively in written and spoken English.
