*Role Description*

The Cancer Immunotherapeutics Unit within Pfizer Vaccines Research and Development seeks to discover novel therapeutic vaccines for the treatment of cancer using a variety of innovative vaccine technology platforms in combination with immunomodulatory agents. The multi-component therapeutic vaccines being developed, including recombinant viral vaccine vectors, oncolytic viruses, and nucleic acids in combination with various immunotherapy agents, offer the potential to revolutionize cancer therapy and may provide new hope to patients worldwide.We are seeking an experienced Scientist to join the Molecular and Cellular Biology group in La Jolla, CA. As a member of a diverse, multi-disciplinary team, the Scientist will provide critical molecular virology expertise to advance novel cancer therapeutics from discovery through pre-clinical studies and into the clinic. The Scientist will support the design, engineering, production, purification, and characterization of virus-based cancer therapeutics. The Scientist will utilize established, as well as optimize and develop additional modern molecular virology techniques, to deliver viral components of vaccine or oncolytic-based immunotherapy regimens.The successful candidate will have considerable experience with various molecular biology techniques, including cloning by conventional, recombination-based, or other related techniques. The Scientist must be proficient in using DNA analysis software packages such as DNAStar and/or VectorNTI. The candidate must have experience with aseptic technique, mammalian cell culture, and handling of Biosafety Level 2 (BSL2) agents. The Scientist will be required to work with BSL2 infectious viruses such as Vaccinia Virus and Adenovirus. Experience with virus engineering, production, purification, and characterization, as well as CRISPR or synthetic biology, are highly desirable. The candidate will work in a laboratory-centered role with limited to moderate supervision. As a member of a multi-disciplinary team, the candidate will be accountable for delivering high quality vaccine components for pre-clinical studies in a timely manner, independently performing, troubleshooting, and rigorously documenting experiments, as well as critically evaluating and presenting results to discipline and project teams. A major emphasis is placed upon laboratory cross-training to increase the scope of practical scientific abilities, as well as the theory behind such practical work.


Key responsibilities are laboratory based (80-90%) and include, with limited to moderate supervision:* Genetic engineering of recombinant viruses (insertion of foreign transgenes, deletion of viral genes) by established molecular virology methods * Research scale production, purification, and characterization of recombinant viruses* Maintenance of mammalian cell culture lines in adherence with proper aseptic techniques* Cloning of genes using traditional, recombination based, and/or other standard molecular biology techniques* Independently designing, executing, and troubleshooting laboratory experiments with rigorous documentation in electronic lab notebooks* Analyzing, summarizing, and presenting data to work groups and project teams* Strictly adhering to established environmental health and safety guidelines, including work with BSL2 viruses


* B.Sc. or M.Sc. in life sciences, preferably in molecular biology / virology / cellular biology * At least 5 years (M.Sc.) or 6 years (B.Sc.) working in a laboratory in an academic or (preferably) industrial setting* Required technical skills/experiences: Mammalian cell culture, PCR, cloning, DNA sequence analysis, DNA purification, DNA transfection, handling of BSL2 or greater viruses, effective written and oral communications* Desirable technical skills/experiences: Virus genetic engineering, virus production/purification, virus infectivity assays, virus characterization techniques, cell-based assays, qPCR, western blotting, ELISA, recombination-based cloning, CRISPR-Cas9, synthetic biology techniques, ultracentrifugationAdditional requirements:* Ability to perform standard laboratory operations while sitting/standing* Work with Biosafety level 2 infectious viruses* Ability to perform mathematical calculations and complex data analysis* No/infrequent travel required* Must receive and maintain required vaccinations or provide satisfactory proof of same#LI-SA1
