Description **ROLE SUMMARY**

Pfizer's Vaccine Research & Development (VRD) organization is a recognized leader in the end to end development of vaccines and is responsible for activities from research to clinical development with a focus on the prevention and treatment of high medical-need diseases, delivering innovation that results in breakthrough therapies for patients and contributes significant value to the Pfizer enterprise. VRD priorities are focused on "high medical need, high-value" infectious disease targets for each life segment and active immuno-oncology (cancer vaccine) targets aligned with Pfizer's oncology research and business units.

As one of the strategic pillars of VRD, the Cancer Vaccines & Immunotherapeutics (CVIT) group is primarily focused on cancer vaccine approaches. These are based on active immunization and oncolytic virus based platforms that include combinations with other immunotherapeutic agents for treating various cancers with the aim of increasing both response rates and duration of response along with improved safety profiles as compared with other passive immunotherapy or chemotherapy approaches.

We are seeking an experienced Senior Associate Scientist to join the Molecular and Cellular Biology group of CVIT in La Jolla, CA. As a member of a diverse, multi-disciplinary team, the candidate will provide critical virology and cell-based assay expertise to advance novel cancer therapeutics from discovery through pre-clinical studies and into the clinic. The candidate will conduct virus productivity studies and cell based assays to support the selection and optimization of production cell lines for oncolytic viruses. The individual will utilize established methods and assays, as well as develop and optimize additional techniques suitable for high-throughput screening of cell lines and viruses with diverse properties. Additionally, this Senior Associate Scientist will support the production, purification, and characterization of virus-based cancer therapeutics.

The successful candidate will have considerable knowledge and practical experience of aseptic technique, the culture of mammalian cell lines (adherent and suspension), handling of Biosafety Level 2 (BSL2) agents, and virus/ cell based assays (e.g. reporter, cell viability, virus titer assays). The individual will be required to work with BSL2/BSL2+ infectious viruses such as Vaccinia Virus and Adenovirus in accordance with Pfizers OHW and EHS policies. Experience with virus production, purification, and characterization in a variety of molecular (e.g. PCR, qPCR, WB) and cellular (e.g. plaque, neutralization) assays are highly desirable. Experience of molecular engineering using conventional, recombination-based, or other cloning techniques would be advantageous.

The candidate will work in a laboratory-centered role with moderate supervision. As a member of a multi-disciplinary team, the candidate will be accountable for delivering high quality results in a timely manner, performing, troubleshooting, and rigorously documenting experiments, as well as critically evaluating and presenting results to discipline and project teams. A major emphasis is placed upon scientific and laboratory cross-training to increase the scope of practical scientific abilities, as well as the theory behind such practical work.


Key responsibilities are laboratory based (80-90%) and include, with limited to moderate supervision:

• Virus growth/productivity assessments in variety of adherent and/or suspension cell lines using established assays and/or developing new technologies

• Maintenance of mammalian cell culture lines in adherence with proper aseptic techniques

• Research scale production, purification, and characterization of recombinant viruses

• Independently designing, executing, and troubleshooting laboratory experiments with rigorous documentation in electronic lab notebooks

• Analyzing, summarizing, and presenting data to work groups and project teams

• Strictly adhering to established environmental health and safety guidelines, including work with BSL2 viruses


• B.Sc. or M.Sc. in life sciences, preferably in molecular biology / virology / cellular biology

• At least 3 years (B.Sc.) or 0-3 years (thesis based M.Sc.) working in a laboratory in an academic or (preferably) industrial setting

• Required technical skills/experiences: Aseptic technique, mammalian cell culture (adherent and suspension), cell and/or virus-based assays (reporter expression, cell viability, virus titer), handling of BSL2 or greater viruses, effective written and oral communications

• Desirable technical skills/experiences: Virus genetic engineering, PCR/qPCR, cloning, DNA sequence analysis, DNA purification, DNA transfection, virus production/purification, virus characterization techniques, western blotting, ELISA


+ Ability to perform standard laboratory operations while sitting/standing

+ Work with Biosafety level 2 infectious viruses

+ Ability to perform mathematical calculations and complex data analysis

+ No/infrequent travel required
